Rumen fluid pretreatment promotes anaerobic methane production: revealing microbial dynamics driving increased acid yield from different concentrations of corn straw.

In this work, the corn straw (CS) with concentrations of 3%, 6%, and 9% (w/v) were pretreated by rumen fluid (RF) and then used for batched mesophilic biogas production. The results showed that after a 6-day pretreatment, volatile fatty acid (VFAs) production of 3.78, 8.27, and 10.4 g/L could be found in 3%, 6%, and 9%, respectively. When concerning with biogas production, the highest accumulative methane production of 149.1 mL CH4/g volatile solid was achieved by 6% pretreated CS, which was 22% and 45% higher than 3% and 9%, respectively. Also, it was 3.6 times higher than the same concentration of unpretreated CS. The results of the microbial community structure analysis revealed that the 6% CS pretreatment not only maintained a microbial community with the highest richness and diversity, but also exhibited the highest relative abundance of Firmicutes (45%) and Euryarchaeota (3.9%). This high abundance was conducive to its elevated production of VFAs and methane. These findings provide scientific reference for the utilization of CS and support the development of agricultural waste resource utilization and environmental protection.

PSII-19 Fecal microbiota composition of weaned piglets under mixed management and poor housing conditions fed diets increased in threonine, methionine and tryptophan

Abstract This study evaluated the effects of increased dietary Thr, Met and Trp supplementation on fecal microbiota composition of weaned piglets raised under contrasting sanitary conditions [(GOOD SC) or under mixed management and poor housing conditions (MM+POOR SC)] during the nursery phase. A total of 144 entire male piglets [body weight (BW) = 6.33 ± 0.91 kg] from a high sanitary status (HSS) farm were randomly housed following a 2×2 factorial arrangement regarding BW blocks. Factorial arrangement was composed of two contrasting SC (GOOD or MM+POOR) and two diets [control, with nutritional requirements according to NRC (2012); or supplemented with Thr:Lys, Met+Cys:Lys and Trp:Lys 20% more than the control]. In total, 12 pens with three piglets each were used for treatment. On d -2, the 144 HSS piglets were allocated to one of two facilities under contrasting SCs. Also on d -2, to mimic a mixed management in the MM+POOR SC, 48 entire male piglets, from a commercial farm with low sanitary status (LSS), were randomly allocated by two piglets per pen into the same pens where the three HSS piglets were. On d 0, the two LSS piglets were removed from the HSS pens and allocated as sentinels into the remaining pens (four/pen) located between pens with HSS piglets. The MM+POOR SC had a weekly cleaning routine only to remove the manure located under the slatted floor, while in the GOOD SC, a daily cleaning routine and a high level of biosecurity protocol were adopted. One fecal sample per pen was collected on d 0, 21 and 42. The V4 region of the bacterial 16S rRNA gene was amplified from each fecal sample to evaluate ecological changes into bacterial communities and taxonomy. Mean relative abundance for each family/genus/species (taxon) across groups were depicted as a heatmap using the log2 transformed relative abundance of the most dominant taxa based on a 2% cutoff and network analysis of clusters of co-occurrences of the most relevant taxa (Figure 1). Overall, fecal microbiome analyses revealed no differences (P >0.05) in bacterial community composition and individual taxon abundances at the bacterial genus level across treatments over time. The alpha- and beta-diversity analysis showed no diet or SC differences (P >0.05). Additional community structure analysis based on co-occurrence network mapping revealed the absence of major temporal changes in architecture or keystone nodes (taxa) for HSS animals housed in either GOOD or MM+POOR SCs, despite the AA+ supplementation. The analysis revealed no specific taxon or taxa that were preferentially enriched or diminished over time (P > 0.05). According to our results, the microbial fecal composition of weaned piglets under GOOD or MM+POOR SCs measured using 16S rRNA sequencing analysis was stable including the major central taxa with no Thr, Met and Trp supplementation effect.

Response of the Endophytic Microbial Composition in Amaranthus Roots to Different Fertilization Treatments

To explore how endophytic microbial compositions in amaranth roots are influenced by various fertilization methods and to determine whether these microbes are associated with amaranthin formation, we conducted an analysis of the endophytic microbial community structure. The roots of amaranth plants subjected to different fertilization treatments—conventional fertilization without potassium (NP), conventional fertilization without phosphorus (NK), conventional fertilization without nitrogen (PK), and balanced fertilization (NPK)—were examined. The results showed that the proportions of Streptomyces, Actinospica, and Burkholderia-Caballeronia-Paraburkholderia in the amaranth roots under the balanced fertilization (NPK) treatment were all greater than those in the amaranth roots under the nitrogen (PK), phosphorus (NK), and potassium (NP) deficiency fertilization treatments. In contrast, the proportions of Phenylobacterium, Acrocalymma, Neocosmospora, Fusarium, Acidovorax, Gibellulopsis, Cladosporium, Dactylonectria, and Gibberella in the amaranth roots under the nutrient deficiency fertilization (NP, NK, and PK) treatments were higher than those in the amaranth roots under the balanced fertilization treatment. Additionally, a significantly positive correlation was found between Streptomyces and the amaranthin content. Furthermore, Acrocalymma, Neocosmospora, and Fusarium exhibited significantly negative correlations with the amaranthin content. The above results suggested that endophytes could easily colonize in amaranth roots as beneficial microorganisms under balanced fertilization conditions. In other words, the balanced fertilization (N, P and K fertilizers are 188.0, 53.0 and 50.0 kg·hm−2, respectively) could recruit more beneficial endogenous microorganisms in amaranth roots for improving their growth and quality.

Metagenomic landscape of sediments of river Ganga reveals microbial diversity, potential plastic and xenobiotic degradation enzymes

The Ganga is the largest river in India, serves as a lifeline for agriculture, drinking water, and religious rites. However, it became highly polluted due to the influx of industrial wastes and untreated sewages, leading to the decline of aquatic biodiversity. This study investigated the microbial diversity and plastic-xenobiotic degrading enzymes of six sediment metagenomes of river Ganga at Prayagraj (RDG, TSG, SDG) and Devprayag (KRG, BNG, BRG). The water quality parameters, higher values of BOD (1.8–3.7 ppm), COD (23–29.2 ppm) and organic carbon (0.18–0.51%) were recorded at Prayagraj. Comparative analysis of microbial community structure between Prayagraj and Devprayag revealed significant differences between Bacteroidetes and Firmicutes, which emerging as the predominant bacterial phyla across six sediment samples. Notably, their prevalence was highest in the BRG samples. Furthermore, 25 OTUs at genus level were consistent across all six samples. Alpha diversity exhibited minimal variation among samples, while beta diversity indicated an inverse relationship between species richness and diversity. Co-occurrence network analysis established that genera from the same and different groups of phyla show positive co-relations with each other. Thirteen plastic degrading enzymes, including Laccase, Alkane-1 monooxygenase and Alkane monooxygenase, were identified from six sediment metagenomes of river Ganga, which can degrade non-biodegradable plastic viz. Polyethylene, Polystyrene and Low-density Polyethelene. Further, 18 xenobiotic degradation enzymes were identified for the degradation of Bisphenol, Xylene, Toluene, Polycyclic aromatic hydrocarbon, Styrene, Atrazene and Dioxin etc. This is the first report on the identification of non-biodegradable plastic degrading enzymes from sediment metagenomes of river Ganga, India. The findings of this study would help in pollution abatement and sustainable management of riverine ecosystem.

Emissions of N2O AND CH4 gases and soil bacterial community under integrated systems in the tropical region

The use of integrated production systems is widespread, mainly in tropical areas. This practice could contribute to soil conservation, increasing biodiversity and environmental services while reducing greenhouse gases emissions. The present work assessed bacterial biodiversity and correlations with nitrous oxide (N2O) and methane (CH4) emissions in soils under integrated production systems. Analyses of bacterial diversity and community structure were carried out using DGGE (denaturing gradient gel electrophoresis). Samples for the evaluation of greenhouse gases were collected in the field and measured using a gas chromatograph. The assessed treatments in a rainy and dry season were: crop (L), pasture (P), planted eucalypt forests (F), pasture followed by crop (IPL), crop followed by pasture (ILP), crop with forest (ILF); pasture with forests (IPF); crop with forest followed by pasture with forests, pasture with forests followed by crop with forest and crop, grassland and forest. In the rainy season, most N2O emissions occurred in the ILP with an average flow of 42.33 µg N m−2 h−1, and the lowest flow was observed in the ILF, with an average flow of 5.53 µg N m−2 h−1. In the dry season, higher emission occurred in the IPL, with an average flow of 4.17 ug N m−2 h−1. In this period, we observed gas consumption in F, the highest N2O consumer with a flow of −4.14 µg N m−2 h−1. In both seasons, CH4 was consumed, with IPF as the most significant consumer with an average flow of −14.19 µg C m−2 h 1(dry); F was the most significant consumer with an average flow −12.32 µg of C m−2 h−1. Bacterial community structure was affected by the agricultural production system and the seasons. Methane emissions and pH showed a correlation with bacterial diversity in different treatments. In contrast, nitrous oxide was negatively correlated with bacterial diversity. Based on our results, we suggest that integrated production systems and plant coverage modulate soil bacterial community structure and biodiversity which, in turn, affect soil processes related to higher or lower methane and nitrous oxide emissions.

Temporal and Spatial Dynamics of Plankton Communities in the Lower Haora River, Tripura, India

Planktons are significant bioindicator of ecosystem functioning, knowledge of the seasonal fluctuation in the plankton population in riverine waters of Indian subcontinent is rather limited. In the present study, analysis regarding spatio-temporal variations based on different multivariate statistics and indicator value analysis is done along with analysis of community structure of plankton assemblages. A total of 46 plankton taxa out of which 38 phytoplankton and 8 zooplankton taxa have been identified based on examination of samples taken from four locations over three sampling seasons. The primary phytoplankton families that involve Chlorophyceae, Bacillariophyceae, Cyanophyceae, Euglenophyceae and Composopogonophyceae. Chlorophyceae dominated the phytoplankton population, followed by Bacillariophyceae and Cyanophyceae. The plankton abundance was recorded highest in spring, followed by winter, and the least reported in monsoon season. The high average percent contribution of Aulacoseira, Oscillatoria, and Spirogyra species is well represented through SIMPER analysis. In addition, the Shannon–Wiener index (H) was calculated to determine plankton diversity, and Pielous' Evenness index (E) was calculated to determine individual distribution within a community whereas Margalef’s Richness index showed species richness of the study area. This qualitative and quantitative study will aid future research on the occurrence of plankton as well as the more trustworthy information generation on fish diet and feeding behaviors. The condition of planktons as bioindicators would indirectly provide an estimation of the ecosystem's natural state or amount of pollution.

Early detection of emerging viral variants through analysis of community structure of coordinated substitution networks

The emergence of viral variants with altered phenotypes is a public health challenge underscoring the need for advanced evolutionary forecasting methods. Given extensive epistatic interactions within viral genomes and known viral evolutionary history, efficient genomic surveillance necessitates early detection of emerging viral haplotypes rather than commonly targeted single mutations. Haplotype inference, however, is a significantly more challenging problem precluding the use of traditional approaches. Here, using SARS-CoV-2 evolutionary dynamics as a case study, we show that emerging haplotypes with altered transmissibility can be linked to dense communities in coordinated substitution networks, which become discernible significantly earlier than the haplotypes become prevalent. From these insights, we develop a computational framework for inference of viral variants and validate it by successful early detection of known SARS-CoV-2 strains. Our methodology offers greater scalability than phylogenetic lineage tracing and can be applied to any rapidly evolving pathogen with adequate genomic surveillance data.

Application of K-Core analysis based on complex network community structure to rolling bearing fault diagnosis

A complex network is a method to analyze nonlinear time series on multiple temporal and spatial scales. K-core analysis of several typical nonlinear series was studied, and then based on this, a fast algorithm method was employed to divide the community structure. According to the definition of k-core, the rolling bearings complex network was divided into different cores at the router level, and the main characteristic quantities such as degree distribution and clustering coefficient of every k-core can be analyzed. The results show that there is a corresponding relationship between the characteristic parameters and the community structure of the bearing. The analysis results show that compared with the traditional method, the method of complex network community structure and k-core analysis has a better diagnosis effect.

The promotion of the atrazine degradation mechanism by humic acid in a soil microbial electrochemical system

The enhancing effects of anodes on the degradation of the organochlorine pesticide atrazine (ATR) in soil within microbial electrochemical systems (MES) have been extensively researched. However, the impact and underlying mechanisms of soil microbial electrochemical systems (MES) on ATR degradation, particularly under conditions involving the addition of humic acids (HAs), remain elusive. In this investigation, a soil MES supplemented with humic acids (HAs) was established to assess the promotional effects and mechanisms of HAs on ATR degradation, utilizing EEM-PARAFAC and SEM analyses. Results revealed that the maximum power density of the MES in soil increased by 150%, and the degradation efficiency of ATR improved by over 50% following the addition of HAs. Furthermore, HAs were found to facilitate efficient ATR degradation in the far-anode region by mediating extracellular electron transfer. The components identified as critical in promoting ATR degradation were Like-Protein and Like-Humic acid substances. Analysis of the microbial community structure indicated that the addition of HAs favored the evolution of the soil MES microbial community and the enrichment of electroactive microorganisms. In the ATR degradation process, the swift accumulation of Hydrocarbyl ATR (HYA) was identified as the primary cause for the rapid degradation of ATR in electron-rich conditions. Essentially, HA facilitates the reduction of ATR to HYA through mediated bonded electron transfer, thereby markedly enhancing the efficiency of ATR degradation.

Combined metagenomics and metabolomic analysis of microbial community structure and metabolic function in continuous soybean cropping soils of Songnen Plain, China

Continuous cropping has a negative effect on soybean yield. In this study, a positioning experiment was conducted starting in 2015, with three treatments: maize–soybean rotation (SMR), 2-year maize, 2-year soybean rotation cropping (SC2), and 8-year soybean continuous cropping (SC8). We determined soybean yields (2015–2022) and analyzed soil microbial communities, functions, and metabolites composition in the 0–20 cm tillage layer using metagenomics technology and GC–MS technology during soybean flowering in 2022. Results indicated that continuous cropping (SC8) significantly reduced soybean yield compared to crop rotation (SMR) during the experimental period, while SC8 showed higher yield than SC2 in 2022. Compared to SMR, SC8 significantly increased soil N content and significantly decreased pH and TP, AP, and AK content. However, the pH and AK contents of SC8 were significantly higher than those of SC2. LeFSe analysis showed that Friedmanniella, Microlunatus, Nitrososphaera, Rubrobacter, Geodermatophilus, Nitriliruptor were enriched in SC8. Gaiella, Sphaerobacter, Methyloceanibacter were enriched in SC2. Sphingomonas, Cryobacterium, Marmoricola, Haliangium, Arthrobacter, Ramlibacter, Rhizobacter, Pseudolabrys, Methylibium, Variovorax were enriched in SMR. And the relative abundance of Cryobacterium, Marmoricola, Haliangium, Arthrobacter, Ramlibacter, Rhizobacter, Methylibium, Variovorax was significantly positively correlated with yield, while the relative abundance of Gaiella and Sphaerobacter was significantly negatively correlated with yield. SC8 significantly increased the abundance of genes in nitrogen metabolism and significantly decreased the abundance of genes related to phosphorus and potassium metabolism compared with SMR. However, the abundance of genes in potassium metabolism was significantly higher in SC8 than in SC2. Metabolomic analysis showed that compared to SMR, SC8 decreased the abundance of carbohydrates, ketones, and lipid. However, the abundance of carbohydrates, ketones, and lipid was significantly higher in SC8 than in SC2. Mantel test showed that soil pH and AK significantly affected soil microbial community, function, and metabolite composition. Characterizing the regional distribution, interaction with microorganisms, and sources of dissolved organic matter for summer rainfall: Insights from spectroscopy, community structure, and back-trajectory analyses

Dissolved organic matter (DOM) in rainfall participates in many biogeochemical cycles in aquatic environments and affects biological activities in water bodies. Revealing the characteristics of rainfall DOM could broaden our understanding of the carbon cycle. Therefore, the distribution characteristics and response mechanisms of DOM to microorganisms were investigated in different regions of Hebei. The results indicated that the water quality of the northern region was worse than that of the middle and southern regions. The two protein like components (C1, C2) and one humic like component (C3) were obtained; at high molecular weight (MW), the fluorescence intensity is high in the northern region (0.03 ± 0.02 R.U.), while at low MW, the fluorescence intensity is highest in the southern region (0.50 ± 0.18 R.U.). Furthermore, C2 is significantly positively correlated with C1 (P < 0.01), while C2 is significantly negatively correlated with C3 (P < 0.05) was observed. The spectral index results indicated that rainfall DOM exhibited low humification and highly autochthonous characteristics. The southern region obtained higher richness and diversity of microbial species than northern region (P < 0.05). The community exhibits significant spatiotemporal differences, and the Acinetobacter, Enterobacter, and Massilia, were dominant genus. Redundancy and network analyses showed that the effects of C1, C2, and nitrate on microorganisms increased with decreasing MW, while low MW exhibited a more complex network between DOM and microorganisms than high MW. Meanwhile, C1, C2 had a large total effect on β-diversity and function through structural equation modeling. The backward trajectory model indicates that the sources of air masses are from the northwest, local area, and sea in the northern, middle, and southern regions, respectively. This study broadened the understanding of the composition of summer rainfall DOM and its interactions with microorganisms during rainfall.

Effect of Cucumber Continuous Monocropping on Traditional Chinese Medicine Residue through Analysis of Physicochemical Characteristics and Microbial Diversity

The use of traditional Chinese medicine (TCM) residue as a crop culture substrate has unique advantages in alleviating the obstacles associated with continuous monocropping, such as increasing production, improving quality and alleviating pests and diseases. However, the effect of TCM residue application on substrates in continuous monocropping practices has not been determined. In this study, the cucumber variety “Jinyou No. 10” was used as the material, and fermented TCM residue, vermiculite and perlite were used as organic substrates (3:1:1). The cucumbers were cultivated on substrates for different durations of continuous monocropping, which were the first cropping cycle (A1), second cropping cycle (A2), third cropping cycle (A3) and fourth cropping cycle (A4). The control (A0) was the substrate sample without any crop planted in it. After the cucumbers were harvested, substrate samples (areas around the cucumber roots) were collected. The physiochemical properties of the cultivated substrates were determined, and the microbial community structures were analyzed through 16S rRNA and ITS sequencing. The physiochemical indices of the substrates with different durations of continuous monocropping (A1–A4) were significantly different than those of the control (A0) substrate. Moreover, the continuous cropping of cucumber had greater effects on fungal communities than on bacterial communities. Bacterial community structure analysis revealed a greater proportion of important bacterial taxa (Proteobacteria, Chloroflexi, and Nitrospirae) in the continuous monocropping substrates than in the A0 substrate. For the fungal community, Ascomycota accounted for the largest percentage of the fungal community in all the samples. The diversity of the microbial community was found to be influenced primarily by electrical conductivity, organic matter content, pH and total potassium content according to the correlation analysis of physicochemical properties and relative abundance of the microbial community. Our study would provide a basis for addressing persistent challenges in continuous cropping and for obtaining the utmost benefit from using TCM organic residue waste.

Bacterial community structure analysis of sludge from Taozi lake and isolation of an efficient 17β-Estradiol (E2) degrading strain Sphingobacterium sp. GEMB-CSS-01

Environmental challenges arising from organic pollutants pose a significant problem for modern societies. Efficient microbial resources for the degradation of these pollutants are highly valuable. In this study, the bacterial community structure of sludge samples from Taozi Lake (polluted by urban sewage) was studied using 16S rRNA sequencing. The bacterial phyla Proteobacteria, Bacteroidetes, and Chloroflexi, which are potentially important in organic matter degradation by previous studies, were identified as the predominant phyla in our samples, with relative abundances of 48.5%, 8.3%, and 6.6%, respectively. Additionally, the FAPROTAX and co-occurrence network analysis suggested that the core microbial populations in the samples may be closely associated with organic matter metabolism. Subsequently, sludge samples from Taozi Lake were subjected to enrichment cultivation to isolate organic pollutant-degrading microorganisms. The strain Sphingobacterium sp. GEMB-CSS-01, tolerant to sulfanilamide, was successfully isolated. Subsequent investigations demonstrated that Sphingobacterium sp. GEMB-CSS-01 efficiently degraded the endocrine-disrupting compound 17β-Estradiol (E2). It achieved degradation efficiencies of 80.0% and 53.5% for E2 concentrations of 10 mg/L and 20 mg/L, respectively, within 10 days. Notably, despite a reduction in degradation efficiency, Sphingobacterium sp. GEMB-CSS-01 retained its ability to degrade E2 even in the presence of sulfanilamide concentrations ranging from 50 to 200 mg/L. The findings of this research identify potential microbial resources for environmental bioremediation, and concurrently provide valuable information about the microbial community structure and patterns within Taozi Lake.

Metagenomic Insight into the Effect of Probiotics on Nitrogen Cycle in the Coilia nasus Aquaculture Pond Water.

Recently, probiotics have been widely applied for the in situ remediation of aquatic water. Numerous studies have proved that probiotics can regulate water quality by improving the microbial community. Nitrogen cycling, induced by microorganisms, is a crucial process for maintaining the balance of the aquatic ecosystem. Nevertheless, the underlying mechanisms by which probiotics enhance water quality in aquatic systems remain poorly understood. To explore the water quality indicators and their correlation with nitrogen cycling-related functional genes, metagenomic analysis of element cycling was performed to identify nitrogen cycling-related functional genes in Coilia nasus aquatic water between the control group (C) and the groups supplemented with probiotics in feed (PF) or water (PW). The results showed that adding probiotics to the aquatic water could reduce the concentrations of ammonia nitrogen (NH4+-N), nitrite (NO2--N), and total nitrogen (TN) in the water. Community structure analysis revealed that the relative abundance of Verrucomicrobiota was increased from 30 d to 120 d (2.61% to 6.35%) in the PW group, while the relative abundance of Cyanobacteria was decreased from 30 d to 120 d (5.66% to 1.77%). We constructed a nitrogen cycling pathway diagram for C. nasus aquaculture ponds. The nitrogen cycle functional analysis showed that adding probiotics to the water could increase the relative abundance of the amoC_B and hao (Nitrification pathways) and the nirS and nosZ (Denitrification pathways). Correlation analysis revealed that NH4+-N was significantly negatively correlated with Limnohabitans, Sediminibacterium, and Algoriphagus, while NO2--N was significantly negatively correlated with Roseomonas and Rubrivivax. Our study demonstrated that adding probiotics to the water can promote nitrogen element conversion and migration, facilitate nitrogen cycling, benefit ecological environment protection, and remove nitrogen-containing compounds in aquaculture systems by altering the relative abundance of nitrogen cycling-related functional genes and microorganisms.

Induced domestication of humic reduction-denitrification coupled bacteria improved treatment of sediment: Performance, remediation effect, and metabolic mechanisms

The high organic matter in river sediment primarily induces black and odorous rebound. Traditional humic-reducing bacteria demonstrate relatively single metabolic functions and restrain the remediation within complex sediment environments. In addition, Ca(NO3)2 is commonly utilized in synergistic with bioremediation to improve the reducing environment of sediments. In this study, a multifunctional bacterial community with humic reduction-denitrification coupled bacteria was domesticated by the step-feeding strategy in an anaerobic baffle reactor (ABR). The performance, remediation effect, and metabolic mechanisms were analyzed. The results indicated that humic-reducing bacteria (HRB) and denitrifying-humic-reducing bacteria (DF/HRB) have quinone-reduction and denitrification capabilities. The synergistic effect of DF/HRBs and Ca(NO3)2 was superior to HRBs and Ca(NO3)2 on the removal of total organic matter(TOM). Microbial community structure analysis revealed an enhanced relative abundance of denitrification and humic-reducing bacteria (e.g., Thauera, Pseudomonas, Sulfurospirillum, Desulfovibrio, Geobacter) in the DF/HRB, resulting in a superior synergistic effect of DF/HRBs with Ca(NO3)2. This work helps to present an innovative approach to domesticate humic-reducing bacteria suited for the remediation environment effectively. It also expands the application of humic-reducing bacteria for in-situ anaerobic remediation of river sediments.

Prebiotic inulin ameliorates SARS-CoV-2 infection in hamsters by modulating the gut microbiome

Current treatment options for COVID-19 are limited, with many antivirals and immunomodulators restricted to the most severe cases and preventative care limited to vaccination. As the SARS-CoV-2 virus and its increasing variants threaten to become a permanent fixture of our lives, this new reality necessitates the development of cost-effective and accessible treatment options for COVID-19. Studies have shown that there are correlations between the gut microbiome and severity of COVID-19, especially with regards to production of physiologically beneficial short-chain fatty acids (SCFAs) by gut microbes. In this study, we used a Syrian hamster model to study how dietary consumption of the prebiotic inulin affected morbidity and mortality resulting from SARS-CoV-2 infection. After two weeks of observation, we discovered that inulin supplementation attenuated morbid weight loss and increased survival rate in hamster subjects. An analysis of microbiome community structure showed significant alterations in 15 genera. Notably, there were also small increases in fecal DCA and a significant increase in serum DCA, perhaps highlighting a role for this secondary bile acid in conferring protection against SARS-CoV-2. In light of these results, inulin and other prebiotics are promising targets for future investigation as preventative treatment options for COVID-19.

Analysis of Microbial Diversity and Community Structure of Rhizosphere Soil of Three Astragalus Species Grown in Special High-Cold Environment of Northwestern Yunnan, China.

Astragalus is a medicinal plant with obvious rhizosphere effects. At present, there are many Astragalus plants with high application value but low recognition and resource reserves in the northwestern area of Yunnan province, China. In this study, metagenomics was used to analyze the microbial diversity and community structure of rhizosphere soil of A. forrestii, A. acaulis, and A. ernestii plants grown in a special high-cold environment of northwestern Yunnan, China, at different altitudes ranging from 3225 to 4353 m. These microbes were taxonomically annotated to obtain 24 phyla and 501 genera for A. forrestii, 30 phyla and 504 genera for A. acaulis, as well as 39 phyla and 533 genera for A. ernestii. Overall, the dominant bacterial phyla included Proteobacteria, Actinobacteria, and Acidobacteria, while the dominant fungal ones were Ascomycota and Basidiomycota. At the genus level, Bradyrhizobium, Afipia, and Paraburkholderia were the most prevalent bacteria, and Hyaloscypha, Pseudogymnoascus, and Russula were the dominant fungal genera. Some of them are considered biocontrol microbes that could sustain the growth and health of host Astragalus plants. Redundancy analysis revealed that pH, TN, and SOM had a significant impact on the microbial community structures (p < 0.05). Finally, triterpene, flavonoid, polysaccharide, and amino acid metabolisms accounted for a high proportion of the enriched KEGG pathways, which possibly contributed to the synthesis of bioactive constituents in the Astragalus plants.

US Nationwide Multi-City Media Coverage of COVID-19 Responses: Community Structure Theory, Belief System, and a “Violated Way of Life”

Community structure analysis compared city characteristics and newspaper coverage of state/local government responses to COVID-19 in 25 major U.S. cities, sampling all 250+ word articles from 4/4/20 to 7/6/20. The resulting 588 articles were coded for “prominence” and “direction” (favorable/unfavorable/balanced-neutral coverage), then combined into each newspaper’s composite “Media Vector” (range=0.3552 to −0.5197, or 0.8749). Twenty-one of 25 newspapers (84%) displayed unfavorable coverage of local COVID-19 responses. Pearson correlations and regression analysis confirmed a muscular “violated way of life” pattern, when a community perceives itself as threatened by a “biological threat or a threat to a cherished way of life.” Political and belief system polarization (in particular percent Evangelical and percent voting Republican) were strongly associated with unfavorable coverage of local pandemic responses, compared to more favorable responses linked to percent voting Democratic or percent Catholic. Vulnerability (percent uninsured) was also linked to negative coverage. Conversely, two different measures of access to healthcare (percent municipal spending on health and welfare, and physicians/100,000) were significantly linked to favorable coverage of the same local government efforts. Community structure theory’s grass roots “bottom up” expectations linking community demographics to variations in reporting on critical issues were robustly confirmed.

Medium optimization and dust suppression performance analysis of microbial-based dust suppressant compound by response surface curve method.

At present, microbial dust suppressants based on microbial communities lack necessary systematic analysis of factors affecting dust suppression performance. Therefore, in this study, the response surface curve method was used to optimize the culture conditions for enrichment of urease-producing microorganisms from activated sludge. The results indicated that when urea = 9.67gL-1, NH4Cl = 5.21gL-1, and pH = 9.57, the maximum urease activity of urease-producing microbial community (UPMC) was 8.22mMmin-1. The UPMC under optimized culture conditions reached a mineralization rate of 98.8% on the 1st day of mineralization. Ureolysis is one of the biological mechanisms that trigger microbial mineralization with the consequent effect of dust suppression. The analysis of microbial community structure indicated that the urease-producing bacteria Sporosarcina sp. had the highest abundance at the genus level in the microbial-based dust suppressant compound. Jeotgalicoccus sp. plays an important role in improving and maintaining the stability of urease. In addition, the optimal UPMC had low pathogenicity, which is extremely attractive for the safe application of microbial dust suppressants.

Paleoecology and paleobiogeography of the latest Miocene site of Shuitangba, Zhaotong, China

Apart from northern and central China, the fossil record of the latest Miocene and Early Pliocene of Asia is not well documented and the record of South China during this interval is especially poor. Shuitangba, a site in Yunnan Province, offers a rare window into the paleoenvironment of the latest Miocene in southwestern China. Over 2400 vertebrate and macrobotanical specimens have been recovered from the site. The faunal assemblage is dominated by aquatic avian taxa and many of the mammalian taxa are those that indicate densely vegetated, water-margin habitats. Pollen and carpological remains indicate a temperate to subtropical, broad-leaved/coniferous forest around standing water, with more open areas containing grasses and herbs. Analyses of clay minerals, chemical weathering, and enamel stable isotopes suggest that Shuitangba was warmer and more humid than today, possibly with more pronounced seasonality. Results of community structure analysis indicate that the Shuitangba mammalian community was different from those of other Late Miocene Chinese sites in the high proportions of aquatic-dependent mammalian taxa. While Shuitangba shared mammalian faunal elements with other Late Miocene sites in Yunnan, it was still faunally distinct. Further, Shuitangba was depauperate in its mammalian fauna, which may have been a result of the depositional setting rather than a true reflection of regional faunal diversity.
